In the latest issue of American Scientist there is an article by Tony Rothman questioning ##E=mc^2##. Are the assertions made in the article valid? Are there comments by other physicists available?

I don't subscribe to the magazine, and there doesn't seem to be much of substance in front of the paywall. Could you perhaps provide a summary of his argument?

The main point of the author was the development of E=mc^2. Before 1905 there were papers with E=3/4mc^2. The author described them in detail and speculated whether there was a problem at high energies with Einstein's formula.
