    Hi can somebody explain to me why cloning long oligonucleotides into vector is difficult?

    I'm not too clear on your question. To insert a sequence into a vector isn't that hard, as long as you already have the necessary amount of the desired sequence on hand. What's difficult is to synthetically assemble a very long nucleotide sequence. Is that what you're asking about?

    It may be tricky because long oligos will have huge melting temperature ... and same for the hybridation/annealing temp.
    So your PCR cloning of long oligos will be if not impossible at least very tricky!
