Hello everybody, this is my first topic here. Nice forum!

I work on yeast and I have a question: I have two proteins, one of the is a G-Protein, they apparently can interact with each other when the G-protein is GTP-bound. I have already demonstrated this with CoIP, but it's better to get an additional result to be more convincing about it. My idea is to purify the two proteins from bacteria and repeat the CoIP in presence or absence of GTP or GDP to see in which condition they interact.

Does anybody of you know this assay? Should I use normal GTP or GTP with an S at the gamma position?

    Thanks for the answers!
