What would happen if an element/some elements of someone's building block DNA was changed/affected in some way? Can building block DNA be affected/changed? (For example, I talk in terms of human improvement here).

What do you men by "element"? If you're talking about altering nucleotides or amino acids, that happens all the time. There are any number of genetic disorders you could examine.

Also, define what you mean by improvement? Altering genetic sequences is not so simple as replacing one section and reaping the benefits. One simple alteration can have widespread consequences.
