    I am working on my undergraduate dissertation and I need to carry out XTT assays. I have a number of queries as I read that different cell lines require different colourant concentrations and incubation times.

    I am working on: HeLa, COLO 679, HaCaT and cord blood cells. It is known that melanocytes and keratinocytes have a low metabolic activity; therefore COLO (melanocytes) and HaCaT (keratinocytes) require a higher concentration of cells, colourant and incubation time. If I have to compare the cell viability of the above four cell lines, shouldn't I be keeping the cell and colourant concentrations and incubation periods constant?

      Lizzina, this is more of a general biology chat forum. There are some of us who work in laboratories but this may not be the best place to find a quick answer to your question.
