I am working on the collagen types. I want to observe them in-vivo with a fluorescence microscopy. What is the spectrum of the collagen types. I couldn't it find on internet. I have to determine the correct filter of the fluoroscopic microscopy, than i should observe the collagen in a gel. I just want to see the fluorescence reflection of the collagen (bright blue light). Does anybody have an idea? Thanks...

Have u tried immunofluorescence assays or is that not suitable for this?

Is this a physiological/biochemical study? Can u just tell me a bit more on what you're looking in to/trying to do/see?

Using any other chemical is not suitable. Right now i am workin in-vitro but in the future i will work in vivo (human), so dying or using assay is not suitable. I am preparing a gel (Agaros) and making a one layer collagen level, than observing under the fluorasanmicroscop. Chosing the correct filter for the re-emitting light is important for me

I don't know much about this but maybe you can search for this article:

Imaging Coronary Artery Microstructure Using Second-Harmonic and Two-Photon Fluorescence Microscopy, Aikaterini Zoumi, Xiao Lu, Ghassan S. Kassab, and Bruce J. Tromberg, Biophys J. 2004 October; 87(4): 27782786.

They visualized collagen and elastin using second-harmonic generation and two-photon excited fluorescence..

      Hope it helps.. :-D
