am staining some cells with antibodies for FACS analysis.
my primary ab conc is 0.2ug/ul.

In one tube i have 1,000,000 cells (the approx volume of these cells is 40ul).i add 1ul of my primary ab to these cells and run my facs
If i have 10,000,000 cells (which is say 100ul of cells). How much of my antibody (ul) should i add to my 10,000,000 cells.

Is it 10ul that i add. Do i need to take into account the volume of my cells?

What is actually important is the number of target receptors on each cell, to provide enough antibody to be sure that every target cell is bound.

There should be a technitian available to answer your questions, I'd try calling whoever usually operates the FACS.
